Exploring Supramolecular Modulation of Polymyxin-Lipopolysaccharide Interactions
Abstract: Polymyxins remain an indispensable drug to treat life-threatening multi-drug resistant Gram-negative infections. To address the growing global challenge of antibiotic resistance, it is key to optimise the pharmacological profile of these antimicrobial lipopeptides and explore potential drug delivery approaches that enhance their performance. We have developed supramolecular polymyxin-based nanostructures formed either by self-assembly or by polymer co-assembly. In this experiment we want to test how these novel PmB formulations affect the interaction of the antimicrobial with models of the Gram-negative outer membrane, the natural target of these drugs. The proposal joins expertise from two projects, one developing deuterated bacterial model membranes mimicking sensitive and resistant bacteria for antibiotic research and one developing novel antibiotic formulations to tackle the spreading antibiotic resistance
