Application of new experimental gaskets to mimic pharmaceutical tabletting
Abstract: Tablet manufacturing involves compressing powder blends into solid compacts, a process that can induce phase transformations in active pharmaceutical ingredients (APIs), potentially affecting drug performance. This proposal outlines a neutron diffraction study using a novel gasket assembly on the Pearl instrument to simulate pharmaceutical tabletting on neat active pharmaceutical ingredient powders. Hydrochlorothiazide (HCT) is selected as the model compound due to its recoverable high-pressure phase, previously observed under compression and confirmed via electron diffraction and synchrotron studies. The new gasket design mimics tablet press geometry and accommodates larger sample volumes, enabling in-situ structural analysis under both pseudo-hydrostatic and non-hydrostatic conditions. The experiment aims to quantify phase transformation extents across pressure ranges up to 3 GPa, providing critical insights into compression-induced polymorphism and enhancing the relevance of high-pressure crystallography to pharmaceutical applications.
