Probing enzyme-induced structural reorganization in lipid nanoparticles

Abstract: Phospholipase D (PLD) is a key enzyme in humans, cleaving the headgroup of phosphatidylcholine lipids to generate phosphatidic acid, thereby regulating lipid metabolism in cells. PLD is also overexpressed in certain cancer types, and in regions of the cell (endosomes) where drug delivery takes place. Our group is investigating the impact of lipid nanoparticle (LNP) structure on PLD activity to engineer nanoparticles to selectively release their cargo. A key challenge in such structure function studies is to decouple the effects of nanoparticle structure and composition, since the structure is often tuned by altering the lipid composition. Recent work demonstrated that stable LNPs of inverse hexagonal (HII) and inverse micellar (Fd3m) phases can be obtained by tuning the ratio of lipid components. This enables us to study how PLD interacts with different LNPs sharing the same lipids - but different molar ratios - and how this interaction drives lipid reorganization within the LNPs for the purpose of exploiting this for drug delivery. The key goal of this experiment is to investigate how PLD activity on LNPs impacts their structure and re-arrangement of their lipid components. Understanding how PLD impacts lipid reorganization in LNPs is key for engineering enzyme responsive LNPs for delivery.
