Structural basis of SorLA in vesicle complexes: towards characterization of Parkinson?s Disease biomarkers

Abstract: Parkinson's Disease (PD) is a progressive neurological disorder in which discovery and characterization of biomarkers remains a key challenge. The Sortilin-related receptor (SorLA, SORL1) has emerged as a promising candidate due to its role in various processes linked to PD pathology and its mutations identified in PD cohorts. SorLA's structural and functional involvement in the disease is still unclear. This project aims to characterize SorLA structure while bound to extracellular vesicle membranes released from human cells. This approach will provide a physiologically relevant model for studying this transmembrane protein. Small-Angle Neutron Scattering (SANS) will be used to investigate the nanoscale structure of both proteins and vesicles. We propose to study SorLa wild-type and its PD-assotiated clinical variants. The study will also evaluate how calcium concentration and pH factors, relevant to SorLA's calcium-binding capacity and intracellular trafficking, affect both wt and mutant SorLA. The expected outcome is a set of structural parameters describing both protein conformations and vesicles architectures, including potential mutation-driven changes, e.g. impaired dimerization or modified membrane affinity. Hydrogen/deuterium contrast variation will be applied to distinguish lipid and protein contributions to the scattering. This work will ultimately enable a clearer understanding of the role of SorLA in PD mechanisms.
