Combining Self Assembled Floating Supported and Protein Tethered Membranes: The Best of Both Worlds for Facile and Accurate Membrane Models
Abstract: Integral membrane proteins (IMPs) are key class of biomacromolecules which is evidence by the fact they make up 60% of current drug targets. Developing facile and robust methods to analytically study this class of proteins is therefore important to enhance our understanding of biochemical pathways including this protein type. We have been developing two approaches to enable the study of IMPs within their native lipid matrix. One, a protein tethered membrane system and, another, a self-assembled floating membrane system. Here we propose examining a mechanism for the production of high protein density, orientated self-assembled floating membranes adjacent SAM coated gold surfaces using a combination of both methodologies. If successful this experiment will demonstrate a new surface technology for the study of IMPs and find use in a number biochemical and biotechnological studies.
