Development of Warfarin-Loaded Polyelectrolyte Complex Nanoparticles of Chitosan and ?-PGA
Abstract: Warfarin, a vitamin K antagonist which is mainly used in thrombosis treatment, has a narrow therapeutic window, variable bioavailability, and instability due to pH sensitivity, extensive biodistribution and unpredictable absorption, thus causing strict monitoring1-4. Therefore, a controlled and stable delivery system in a polyelectrolyte complex (PEC) comprising chitosan (CS, biocompatible polycation) and -polyglutamic acid ( -PGA, polypeptide polyanion) is developed to produce stable nanoparticles with balance surface charge, where amine groups (-NH3+) of chitosan interact with carboxylate groups (-COO-) of -PGA to encapsulate warfarin with the polymer matrix5-8, as illustrated in Figure 1. The nanoparticles stability depends on pH, ionic strength, and chitosan : -PGA ratio, making structural characterisation essential. Although PEC-based nanoparticles effectively protect drugs, enhance solubility and enable controlled release, the exact localisation whether entrapped in the core or interacts with the outer layer- and the entrapment efficiency of the system remain unclear8. Therefore, Spin-Echo Small-Angle Neutron Scattering (SESANS) is proposed to be conducted to analyse the chain conformation, interchain distance, and internal nanostructure by exploiting the contrast to confirm intra- and intermolecular interaction within CS/ -PGA-warfarin complex. The PEC droplet data will be analysed using a log-normal sphere model, which represents the PEC system in solution.
