Unraveling the role of Myelin Basic Protein in maintaining membrane asymmetry in the Myelin sheath
Abstract: Myelin constitutes the membrane material surrounding axons and consists of alternating extracellular and cytoplasmic leaflets, where there is distinct asymmetry in the lipid composition of each leaflet. The bilayers are in close contact providing a low dielectric constant through the compact bilayers, which is essential for efficient and fast saltatory propagation of nerve impulses. Structural changes of the myelin sheath are signatures of several inflammatory neurological disorders. The Myelin Basic Protein (MBP) is one of the main proteins in myelin and plays an essential role in stabilising and maintaining the myelin structure. It is located only between the 2 cytoplasmic faces, where it acts as an intermembrane adhesion protein to maintain the close contact between membranes in the myelin sheath. Our previous experiments at ISIS have revealed that membrane asymmetry of supported Myelin bilayers is lost upon heating the membrane to physiological temperatures, and that this asymmetry is essential for efficient MBP interaction. Here, we propose to utilise neutron reflectometry and its unique sensitivity to differentially isotopically labelled asymmetric SMBs to investigate the role that MBP itself has in maintaining the the asymmetry of the myelin membranes at physiological temperatures, and the role of cytoplasmic-facing anionic lipids in MBP recruitment.
