G-quadruplexes' chemical heterogeneity at the nanoscale probed by SANS
Abstract: Aqueous solutions of guanosine-5-monophosphates (GMP) ? known to form G-quadruplexes made of stacked quartet ? can be induced to turn into high water content hydrogels by the addition of guanosine (Gua). A combination of SAXS, Light Scattering and AFM suggested that Gua/GMP hydrogels are microscopically heterogeneous, formed by Gua-rich disordered microcoils of intertwined flexible filaments (??knots??) connected by GMP-rich long linear threads. To best design the characteristics of the hydrogel depending on the desired biomedical or biotech application, an accurate insight into the molecular structure of the 3D G-hydrogel needs to assess the presence of chemical heterogeneity at the nanoscale. We then propose to perform contrast variation SANS experiments as a key technique for deriving the distribution of the Gua and GMP components in the G-quadruplexes.
