Using SANS2D to resolve the opening of MscL clusters in proteoliposomes in response to challenge by lyso-PC and pexiganan
Abstract: In bacteria, mechanosensitive ion channels such as MscL protect cells against osmotic shock. Although crucial for protecting cells, as with all ion channels, they have the potential to act as an Achilles heel, by providing a target for antimicrobial treatments. It is thought that these work as pressure release valves and respond to an increase in membrane tension by opening. Our previous SANS measurements indicate that these channels cluster into groups of 5 and some preliminary evidence that the addition of the single-tailed lipid, lyso-PC, causes an increase in the dimensions of these clusters, that could be explained in terms of the increase in diameter of the channels when they open. In this experiment, we will use smaller proteoliposomes that will enable SANS to provide a more complete determination of the structure, allowing us to resolve both channel opening and bilayer insertion.
