Understanding the skin barrier design: identifying the conformation of ceramide EOS within a simple Stratum Corneum lipid model

Abstract: The stratum corneum layer of the skin acts as a barrier between the body and the external environment. This is achieved by dead corneocytes encased in a lamellar lipid matrix, which provides the only continuous pathway through the barrier. Skin diseases occur primarily due to alterations in the lipid matrix, which in turn compromises the barrier capability. To better investigate the lipid matrix a range of model systems have been developed that can mimic the structure and permeability behaviour of stratum corneum, however the lipid arrangement in these models remain unknown. For this experiment we have obtained some partially deuterated ceramide EOS, and we aim to determine if the ceramide is arranged either linearly or as a hairpin. This experiment will provide key information on how the lipid unit cells are bound together, providing further insights into the barrier structure.
