Effect of Cholesterol on NHE1 intracellular domain adsorption on lipid bilayers
Abstract: The Na+/H+ exchanger isoform 1 (NHE1) is a transmembrane protein with the primary function of regulating the pH in the cytosol by exchanging one proton with one Na+ ion. The NHE1 intracellular domain (ICD) is an intersically disordered proteins which regulates protein activity via binding of other proteins, as well as by interacting with the anionic phospholipids in the membrane. Although a specific lipid interaction domain (LID) in NHE1/ICD (named here after NHE1/LID) was identified, a detailed picture of the mechanism of NHE1-lipid interaction is still missing. The investigation of NHE1/LID interaction with lipid membranes is part of a larger project, which aims at the characterization of ICD-lipid membrane co-structures. Here we propose to perform Neutron Reflectometry (NR) experiments to investigate how cholesterol can affect NHE1/LID adsroption on lipid membranes.
